QYM/SURFTECH BCIP/NBT Blue One Component Membrane Substrate is ready-to-use and should be equilibrated to the reaction temperature before application.
Chromogenic Reaction Procedure (Western Blot):
1. Wash: After alkaline phosphatase (AP) conjugate incubation, wash the membrane 3 times with TBST washing buffer.
2. Chromogenic Reaction: Add 1 mL substrate and incubate at 20-37 C until bands appear dark blue.
3. Stop: Wash 2-3 times with 1.5 mL deionized water to terminate the reaction before background color develops.
4. Detection: Analyze bands by visual inspection or by imaging to quantify gray-scale values.
